Description : IC50: N/ASennoside A is a dianthrone glycoside isolated from Rhei Rhizoma and senna leaf. Sennoside A and B have identical molecular weights and formulae. Sennosides were known as laxatives causing purgative actions through the biotransformation of rhein anthrone. Sennoside A was reported to have regionally differential effects on spontaneous contractions of colon.In vitro: The gastroprotective activities of sennoside A was found to be due to the up-regulation of PGE2 and the inhibitions of H+/K+-ATPase activity. When measured the H+/K+-ATPase activities of sennoside A, the positive control exhibited inhibitions of 41.1% and 42.4% at 50 M and 100 M, whereas sennoside A showed dose-dependent inhibitions of 17.3% and 27.1% at 50 M and 100 M [1]. In vivo: In a rat model, sennoside A reduced gastric juice, total acidity and increased pH, indicating that proton pump inhibition reduces gastric acid secretion. Furthermore, sennoside A increased PGE2 in a concentration-dependent manner. Moreover, in the intestinal transporting and gastric emptying rate experiment, sennoside A was found to accelerate such GI motility. These results suggested sennoside A possessed significant gastroprotective activities and it might be useful for the gastric disease treatment [1].
